Question
I have a block that is on layer 0 but has a center line included in the block with a Linetype of Center and a Color of Red. When I am in Paper spaces and change the layer color in a viewport with the block to black the block turns black except the center line, it stays red. Can I change the color of the center line to black with out changing any of the blocks properties?

By hard coding an object in a block definition to a colour other than bylayer or byblock, you are disabling the ability of the object to be easily changed in a layout.

In your case, the idea of having the object colour RED on layer 0 implies that you want it RED everywhere, all the time.

The only way I can think of, aside from redefining the block, is to set red to plot as black in your layout ctb ot stb file.
