Why is sterile skim milk used in casein hydrolysis?

Hi Candace:  Thanks for your question.

Casein is the protein in milk, so it provides a source of nutrients for the bacteria and is used to detect the casein hydrolysis enzymes.

Are you asking why skim/whole milk is used?  I think it's because we can get powdered skim milk to make the medium to grow the bacteria.  Also skim milk powder is more stable without the milkfat.
