Question
How do I calculate the field of view for 400x objective? After I have calculated FOV how do I determine lenght of an individual cell

Answer
If you have measured the width of the field all you have to do is estimate how th cell fits into the  field.  For example lets say that at 400x the diameter of the field is 100 microns and cell fills 3/4 of the field it then has a diameter of 75 microns
