Question
Hypothetical question.  If I am studying a protein which is localized to the center of the cell, when I treat it with a growth factor. I want to prove that the protein is entering the nucleus. What kind of imaging will show the protein is in the nucleus not just above or below it in the cytoplasm.

Answer
You have to make z-section of the fixed cells in which nucleus is stained with DAPI. Labeling your protein with fluorescent antibody and looking for co-localization with the nucleus can tell if the protein indeed enters the nucleus.
