Disease is a manifestation of biological organisms.  You have to know the enemy.

A hypothesis is an educated guess based upon some known facts.  A prediction is basically the same though perhaps more of a guess. A theory is a conclusion based upon repeated testing

They are both extensions of the nuclear membrane.  The rough ER acts as the base for ribosomes and the smooth ER does not.  The rough ER is the template for protein synthesis.  The smooth ER is the site of lipid synthesis
