Question
Dear Dr.Skett,

I´m using PEG 400 90% and DMSO 10% as a vehicle, due to problems regarding drug solubility  (subcutaneous administration, 250 uL, in mice). Could you please answer me if PEG 400 is well absorbed by subcutaneous route? I expect that the drug reaches high plasma levels at 45 minutes to 1 hour post administration. Thank you in advance for any help.

Answer
Rodrigo,

Thanks for your question.

I would doubt that much of the PEG 400 is taken up - I assume you mean into the plasma.

DMSO certainly is and rapidly but the chemical nature of PEG 400 would suggest that this is not absorbed.

Is there any indication that it is?

Best wishes,

Paul
