I had a ball drilled a few days ago and the guy drilled the spin spot or max bias out for my thumb.  Does that mean anything or will it change the reaction of the ball?

The Mass Bias is a secondary axis of the weight block of your ball. It is not a problem to drill it out (actually better to take it out completely, then get real close).

So, No. Hopefully, positioning the mass bias near your thumb is helping create the reaction you desired from the ball.
