Question
If I were to move my thumb hole more towards my index finger instead of behind my middle finger, would that help my ball hook more or would it decrease it.

Answer
It will probably not change your reaction at all.  It may change the feel of your grip.  You will only be changing the relationship of the pitches between your thumb and fingers or making the span on your ring finger longer.

If  you want to try and increase the hook potential of your release,  you will need to look for a way to get your fingers below the equator of the ball during the release, much like cupping the ball.
