Question
When I was young, I remember pulling from the racks a huge over-sized horror comic which might of been from the late 70'early
eighties.
Story was about vampires being killed off in a inquisition. Some survive and are lead by a giant sized vampire who bites off the priest's head who's trying to stop them.
Still vivid in my head after all these years. Not sure If it was black or white or color, It was an anthology of tales and this was the most gruesome of the three.

Sorry I don't recognize this story.  Based on your saying it was an over-sized horror comic and based on the level of violence you describe, it was probably black-and-white.

The biggest publisher of black-and-white horror comics in that time period was Warren, which published Creepy, Eerie and Vampirella.  There were a few other publishers that put out a few issues in the earlier 70s, but most had died out by the late 70s.  Marvel did put out a few black-and-white, magazine-sized comics in the time period you describe and these magazines were more violent than their regular comics, but this story sounds likely too violent to have been in a Marvel issue.
