Answer
Dear Paolo:

Which of the following sentences is correctly punctuated?

1) His dad's girlfriend Brandi checks in to see how he's doing.

2) His dad's girlfriend, Brandi, checks in to see how he's doing.

If both are possible, do they have the same meaning?

*** The second sentence is correct.  The first is not.  His dad has only ONE girlfriend.  Giving her name is additional information that is an appositive.  The name is enclosed with commas.
