you know I always suggest to put yourselves into the hands of professional tour guides/drivers, but in this case if you are willing to be driving by yourselves and you are not scared about our roads, you could catch the road 222 from Florence to Siena and drive through the vineyards in the wine area. From the exit of Florence follow the BLUE signs for Siena (the green signs will lead you to the 4 lanes road that is also panoramic but bot as the old road. On the old road you can stop in Greve in Chianti, Panzano, Radda, Castellina and some other villages of the area. If you instead would like to make a faster thing you can take the green signs for Siena and you drive on the highway. It's less panoramic but you can make a cpouple of detours and stop in San Gimignano (very touristic) and Monteriggioni. This itinerary is faster and with less winding roads.
From Siena to Rome, instead you can take the SS (now they call it SR in Italy but it's the same thing), N°2 (also known as Cassia) and go all the way South towards Rome. From Siena it's easy, just take the BLU indication to Rome, same thing as Florence-Siena, if you take the green ones you will be led on the motorway. You can consider the idea of stopping and having lunch at the lake of Bolsena and maybe making a stop in the particular village of Civita di Bagnoregio.
