i am doing research work on  polymeric nanoparticles  for ocular drug  delivery.my question is how to fix the dose of  antibacterial drug for ocular .please tell me

Hi Senthil,

There are numerous ways but the way I preferred is to first prepare nanoparticles and then characterize them for drug entrapment efficiency, and drug loading etc.

In this process, you get an idea of how much drug is available per mg of nanoparticles. You can fix the dose then for ex: If in 10 mg  of nanoparticles, you are getting 2mg of drug means that 10mg of nanoparticles suspension in 1 mL of vehicle; the dose availability should be 2mg/ml.

The release of nanoparticles are slow so keep this in mind. Perform a in-vitro release study of nanoparticles and see how much drug is releasing per hour. In antibiotic, MIC (minimum inhibitory concentration) concentration of drug is required to show antibacterial effect. See if that concentration of drug is available in solution.

Next thing will be to go for antibacterial assay in microbial plates to see if the calculated amount of nanoparticles are providing same effect as marketed/simple solution.
