Hi Mark,
I have some very interesting stamps that my Grandfather had given me.
They're from the U.S. Duck Hunting Wildlife Edition, 1934-1961.
My question is, "Some of these stamps have a signature on the front.
Does this signature deplete the value of these certain stamps? They have
never been circulated.

The typical use of a federal duck stamp is for the individual hunter to affix the stamp to a duck hunting license and then sign across the face of the stamp in ink.  Duck stamps are valued either "unused" with no signature and full gum on the back, or "used" with a signature and typically no gum on the back.  Used duck stamps are valued less than unused, and if the hunter managed to damage the stamp by folding it or otherwise harming the paper, then the value is greatly diminished.
