What is the best route to take from Alicante Airport to Valencia Airport and what is the estimated toll cost for the route, one way only.

Hello Alan, sorry for the delay with my answer to your question.
You should take AP7 highway along the coast, tolled, then it changes the name to A7 when it's free of tolls, near Valencia (that P in the name of some higways in spain means Peaje = toll)
Near Valencia you have the following options
Use the route proposed by ViaMichelin, taking V30 ring heading to A3-Madrid, the airport is not far once you take A3, and it's on your right side and well marked.

or
do not enter valencia and follow A7 and then take the exit to A3 heading Valencia, the airport is well marked and in your left side as you approach to Valencia from A3.
There are some km more but this highway is much better than V30 option, this V30 have some stretch with works but it's mostly free of works now and has been renewed. I still find it not very good, but you save some km by using V30.
Whatever option you choose it will take you about 2h and cost 13,90 in tolls according to ViaMichelin.
