Question Hi,
I will be in Cuenca between 30th September-3rd October.
After that, I would like to visit Barcelona.
How do I get to Barcelona? Can i take a train or bus, how far is it and how much does it cost?
Is one night and two days enough for Barcelona?
Then, I need to go to Madrid.
How do i get to from Barcelona to Madrid?
How far is it and how much does it cost?
Is one day enough for Madrid?
How is the weather the beginning of October?

This is a lot of questions and I'm not sure I can answer everything...
I think it's probably much better to go to Cuenca from Madrid, then go to Barcelona, if you can do that... Cuenca is easier to reach from Madrid, and yes, you can take a bus (probably easier) or a train. Check this website: http://www.cuencamagica.com/comollegaracuenca.php
One day could be enough to have a very short glimpse of Madrid. Limit yourself to just a couple of things to do, Museo del Prado and strolling the Old Town for example.
The same applies for Barcelona. Plan carefully your trip and what you wanna see. Buy a Bus Turistic ticket (hop on hop off bus) to move around.
Bus or train will allow you to go from MAD to BCN. That's 600 kms. A night train will save you the cost of a night, but it's not cheap. Bus one way can be some 30€. Check www.alsa.es for buses, and www.renfe.es for trains.
