Question What is the best route to take, and the approximate travel time to sail from Cape May, NJ to Block Island?
Answer I guess the answer depends on the size of the sailboat, the bluewater experience the captain and crew have, what you mean by "best," and whether or not time is a factor.
In a straight line, Block Island is about 210 miles. It is deep water all the way and obviously the shortest and fastetst route, putting you about 30 miles off New York City.
If time is no factor, the "all weather" inlets in New Jersey are spaced about 40 miles apart. From Cape May, head for Atlantic City. From Atlantic City head for Mannesquan Inlet (fast, strong current) or Atlantic Highlands.
From either of the northern inlets in New Jersey, you have to decide whether to go outside or inside of Long Island. Personally, I prefer the outside with numerous inlets to choose from. Keep in mind that Shinnecock can be the "roughest."
From Shinnecock to Montauk and then Montauk to Block Island.
Look at a chart and plan on doing forty miles a day. The trip can be even better if you plan on staying a couple of nights in each port.
